Without the comma, I AGREE WITH YOU would be the object of the verb GUESS, which would be said when said agreement is obvious or the speaker finds themself with no choice other than to agree (for some reason). With the comma, it makes no sense. They are two different sentiments, which would normally be separated by a full stop.
